If I read the question correctly, you are asking whether comics characters with exaggerated noses are usually the subjects of broader humor. Probably yes, there may be an intended correlation on the part of the author for the exaggeration to add to the overall absurdityof the situation, but there are probably also a number of social, historical and cultural reasons for why the nose in particular is emphasized.
The question of "funny" depends on the audience. Perhaps in a more homogeneous audience it is easier to come to a consensus on what is funny. For example, do you find Chief Wahoo funny?

Characters with big noses can also be quite tragic, like Cyrano de Bergerac and Pinocchio. If the question is whether big noses are intrinsically funny, the answer is no. Except perhaps for rhinoplasty surgeons.
